WHOLE-CELL RECORDING OF
HYPOXIA DAMAGE OF HIPPOCAMPUS NEUYON IN NEWBORN RAT BRAIN SLICE

Objective: 20 neurons were recorded
by whole-cell patch clamp, which hippocampus neurons, membrane potential
were observed.
Methods: Hippocampus slice of newborn SD rat
brain, it was incubated at artificial cerebrospinal fluid (ACSF) one hour.
The brain slice was perfused 1-2 ml/min whole-cell recording of was used by
blind insertion.
Results: There is spontaneous discharge when
neuron of hippocamus is quiescence, action potential about 40.96±6.9 mV
.The change of membrane potential and potential value (157.54±31mV/min)
were observed when oxygen was stopped and nitrogen was given. This time, if
oxygen was recovered immediately, membrane potential could recovered and
expressed reversibility impolarizable of membrane. If not, irrevesibility
impolarizable of membrane potential was observed (146.2919mV/min).
Conclusion: Hippocampus neuron of newborn rat
brain is very sensitive for hypoxia injury. The changes of membrane
potential injury is in connexion with hypoxia.
