IRON METABOLISM DISORDER AND ITS REGULATION IN ACD RAT:AN EXPERIMENTAL STUDY

Objective: To investigate the regulatory effect of NO, IRP on the iron metabolism disorder in the ACD rat liver on the basis of establishing the rat model of anemia of chronic disease (ACD).

Method: On the basis of traditional adjuvant arthritis rat model, anemia was induced by injecting Freund adjuvant twice so as to enhance the rat’s immune response; Measure the level of iron, Fn, NO and iNOS mRNA of rat liver of the three groups, in the meantime, we also examined the IRP binding activity and the expression level of IRP-2 mRNA in rat liver.

Result: Different degree anemia was induced by injecting the Freund adjuvant in adjuvant arthritis rat, the level of liver iron content and Fn elevated, which was consistent with the characteristics of iron metabolism disorder in ACD; The degree of arthritic edema and anemia in the rats treated with NOS inhibitor L-NAME was slighter than that of ACD rats; The IRP binding activity in ACD rat liver elevated as compared with that of the control group, expression level of IRP-2 mRNA decreased. NO was related positively with IRP binding activity.

Conclusion: It was successful in establishing the model of ACD rat; NOIRP played an important role in the disorder of iron metabolism in the ACD rat liver.
