THE CLINICAL STUDY ON PRODUCTION AND mRNA EXPRESSION  OF TGFβ IN CHILDREN NEPHROTIC SYNDROME

Objective: In order to investigate the clinical significance of the transforming growth factorβ(TGFβ) production and mRNA expression in children nephrotic syndrome .

Methods: In 7 boys and 11 girls with primary nephrotic syndrome, TGFβprotein was detected by ELISA method in serum  and  peripheral blood monoeuclear cell(PBMC) cultured  liquid. Endotoxin-induced TGFβmRNA expression in PBMC was measured by reverse transcription polymerase chain reaction (RT-PCR) method. 18 case (13 girls and 5 boys, health children) as control group.

Results: The results were expressed as: (1)TGFβ protein level was higher in children with nephrotic syndrome than control group especially in PBMC cultured  liquid, in serum 33.68±17.31 vs 16.35±3.28, p<0.05; in PBMC cultured  liquid 143.88±12.51 vs 26.24±5.34 p<0.01; (2)TGFβmRNA expression increased in nephrotic syndrome children group, 4.85±1.01 vs 0.04±0.01, p<0.05.

Conclusion: Our findings suggest that the TGFβmRNA expression and TGFβmight play an important role in pathological mechanism of children nephrotic syndrome.
