IRON MAGNETIC FRACTIONATION CONCENTRATES HEMATOPOIETIC PROGENITOR/STEM

simple way to remove engraftment-unrelated cells before immunomagnetic
purging or CD34 positive selection to reduce the cost of those procedures.
Methods: Using
sterile carbonyl iron (CI) particles and high-energy samarium cobalt
magnets to remove the phagocytic cells from G-CSF (granulocyte-colony stimulating factor) mobilized
peripheral blood progenitor/stem cells (PBSC). A model system was used to
evaluate the efficacy of neuroblastoma purging. A flow cytometry and a
colony assay were used for CD34+ cell enumeration and colony-forming
unit-granulocyte / macrophage (CFU-GM) evaluation, respectively. Results: After a short period incubation of PBSC
with CI followed by the magnetic removal, cell numbers can be reduced to 34.0%±11.8%
(n = 10, P < 0.00001). CD34+
cell recoveries were 84.3%±50.3%
(n = 10, P = 0.09) with a
2.6±1.5-fold
increase (P < 0.00001). CFU-GM
colony recoveries were 100.0%±37.4%
(n = 7, P = 0.998) with
a 3.0±1.1-fold
enrichment (P < 0.00001). The
efficacy of purging neuroblastoma cells seeded into PBSC (n = 3, P = 0.44) was not compromised by CI
fractionation. Conclusions: Carbonyl
iron fractionation is a rapid and an effective means of concentrating
hematopoietic progenitors / stem cells from PBSC without compromising their activities.
This method warrants further testing on clinical trials of stem cell
enrichment before immunomagnetic purging or CD34 positive selection using
CliniMACS.
