THERAPEUTIC EFFECTS OF
DEXAMETHASONE, AMINOGUANIDINE, AMRINONE ON ENDOTOXIC SHOCK Tang

hospital, Beijing, China Objective: To compare the
therapeutic effects of amrinone, aminoguanidine (AG) and dexamethasone (Dex)
on endotoxic shock. Method: Thirty-five New Zealand
rabbits were randomly divided into five groups. Seven rabbits without using
lipopolyssacharide (LPS) is control. Other groups were intavenously
injected LPS (400ug/kg) in 10 min.When BP fell down 10%, three groups
received AG (30mg/kg iv), amrinone (1mg/kg bolus, followed by 10ug/kg.min
iv), Dex (1mg/kg iv) respectively. The last is LPS group. Results: At 6hrs after injection
of LPS, (1). BP, CO, oxygen dilivery (DO2), oxygen consumption (VO2) NO
were kept on normal level in control. (2) Bp and CO of LPS group decreased
to 47±4.4 mm Hg, 170±42.6 ml/ min; DO2 and VO2
declined to8.1±2.4and 2.7±1 ml/kg.min, NO increased to 160.2±17.5 umol/L.
(3) Treatment of Dex significantly improved BP, CO to 72±58mmHg,
262±42.5ml/ min; DO2 to 12.4±3.1; VO2 to
5.1±1.6ml/kg.min, NO to 99.6umol/L (4)These indicators of amrinone were
improved. Bp is 62.8±8.1mm Hg; CO,197.5±39.5 ml/min; DO2 9.5±1.3
ml/kg.min; VO2 4.1±1.5ml/kg.min, NO148.2±14umol/L. (5) BP is
74.3±9.9mmHg;CO is 247.9±46.9ml/min; DO2 and VO2 are
11.2±1.7 ml/kg.min,5±1 ml/kg.min, NO is 58.2±7.7umol/L in AG. Conclusion: All the three drugs
have theraputic effect on endotoxic shock, but aminoguanidine is more
effective than Dex and amrinone.
