文本框: STUDY ON THE EFFECTS OF NITRIC OXIDE ON THE MCMV INFECTED LIVER AND HEPATOCYTE APOPTOSIS IN MICE

Objective: Study the effects of nitric oxide (NO) on the murine cytomegalovirus (MCMV) infected and hepatocyte apoptosis in mice.
Methods: 1. Thirty BALB/C mice were equally divided into 5 groups: In groups A, B and C, the mice were first developed the model of mice liver infection. The mice in group B and C were given NO synthesis inhibitor L-NAME and accelerator L-Arg, on the second day of MCMV inoculation, every day, respectively. The group D and groupE as blanc and L-NAME control group. 2. The serum NO concetration was determined by using Griess method. 3. The liver tissue sections were obtained to detect the expression of iNOS, bcl-2 and fas mRNA (in situ (in situ hybridization), MCMV (ß – gal staining), the expression of MCMV IE gene, apoptostic index (TDT method) and the apoptosis rate from DNA content (flow cytometry). 4. Statistical analysis: Analysis of variance (F test) was used.
Results: 1. The mice in group B the NO synthesis was inhibited, and the MCMV infection in liver tissue was obviously increased, in contrast, the MCMV infection in group C was significantly decreased as compared with that in group A. 2. The apoptosis was milder, bcl-2 expression higher and fas expression lower in group B than those in group A .In contrast, the apoptosis and fas expression in group C were increased as compared with those in group A.
Conclusion: The study demonstrated that NO took part in the action of anti-MCMV and it may be related with the apoptosis of hepatocyte.
