文本框: THE EXPRESSION OF NF-κB IN PERIPHERAL BLOOD MONONUCLEAR CELLS OF CHILDREN WITH JUVENILE RHEUMATOID DISEASE

Objective:  To study the expression of nuclear factor-kappa B(NF-κB) in peripheral blood mononuclear cells (PBMCs) of children with juvenile rheumatoid disease (JRD), and its relationship with the level of serum interleukin (IL)-8 and rheumatoid factor (RF). To investigate the role of NF-κB in the triggering of JRD.
Methods:  The expression of NF-κB was detected by electrophoretic mobility shift assay (EMSA), and enzyme-linked immunosorbent assay (ELISA) was used to measure IL-8 and RF.
Results:  Compared with normal children and children with JRD in remission stage, the expression of NF-κB in PBMCs of children with JRD in active stage was significant statistically elevated (P<0.05). There was a positive linear correlation between the expression of NF-κB and the level of sorum IL-8, and of serum RF. The expression of NF-κB in PBMCs of children in resmission stage of JRD has no difference with that of the controls (P>0.05). The relationship between the expression of NF-κB in PBMCs and the serum IL-8, and the RF was not existed in children of JRD in remssion stage (P>0.05).
Conclusion:  The elevated expression of NF-κB in PBMCs of children in active stage of JRD has close relationship with the level of the serum IL-8 and RF. Thus, we speculate that the transactivation of transcription through activating NF-κB may be one of the most important ways of the triggering of JRD.
