文本框: THE PROTECTIVE EFFECT OF SHENMAI INJECTION ON HYDROGEN PEROXIDE-INJURED CARDIOMYOCYTES

Objective: To explore the protective effect of shenmai injection (SMI) on hydrogen peroxide (H2O2) –injured cardiomyocytes and compare its effect with vitamin C (VitC).
Methods: The cultured neonatal rat cardiomyocytes were divided into control, H2O2,H2O2+VitC and H2O2+SMI groups, after 24-hour further culture, the cell viability was analyzed by MTT assay, lactate dehydrogenase (LDH), malondialdehyde (MDA) content and superoxide dismutase (SOD) activity in culture media were determined respectively, meanwhile cardiomyocyte apoptotic rate was evaluated by flow cytometry.
Results: SMI and VitC could increase the viability of H2O2-injured cardiomyocytes, decrease LDH and MDA content and keep SOD level in the culture media, SMI was more effective than VitC. In addition, SMI could decrease the apoptotic rate of H2O2-injured cardio-myocytes.
Conclusion: SMI could protect cardiomyocytes from damage iduced by oxygen free radicals and exert a better effect than VitC.It might be used in treatment of myocardial diseases such as viral myocarditis in which oxygen free radicals play an important role.
