Objective: Compare the
spatiotemporal expression patterns of different extracellar matrixes (ECMs)
on vitamin A deficient (VAD) malformed rat embryonic hearts to reveal their
effects on the development of the embryonic heart.
Methods: The rat embryos in
different embryonic days (ED) were divided into control group, VAD group
with cardiac malformation (group 1) and VAD group with normal cardiac
structure (group 2). They were applied for the enzymoimmunohistochemical
staining with antibodies against fibronectin(FN), laminin(LMN),
Vimentin(Vn), collagen IV(CoIV) and intergrin b1(b1). The MC-1407DZ Image
analysis system was used to analysis the results.
Results: In control group, the
spatiotemporal expression of ECMs and their receptor b1 were quite different in
the different parts of the rat embryonic heart, the expression of b1 was mainly similar with
FN. In VAD group 1, only the expressions of Fn and b1 had shown both weak and
delay patterns than which in control group, while other ECMs expressed
normally just like the controls. In VAD group 2, the expressions of ECMs
and b1 showed no different with
the controls.
Conclusion ECMs(mainly FN and LMN)
play a certain mediation role on the development of embryonic heart, but
their role location and role mechanism are different. FN and b1 play an important
mediation in fusion of primary cardiac tube and the development of
interventricular septum and endocardial cushion.
