model by scheduled bone marrow transplantation method

Objective: To
explore the method of establishing scheduled bone marrow transplantation
model of murine, and to supply a stable and efficient murine model for the
experiments in vivo on the protection the bone marrow via mdr1 gene transferring into
hematopoietic cells in chemotherapy.
Methods: mdr1 gene was transferred into hematopoietic
cells of murine bone marrow by the mediation of retrovirus vector. And the
transferred murine bone marrow hematopoietic cells were transplanted into
the murine model that had been irradiated by lethal dose 60Co-¦Ã
rays. Then the expression and function of mdr1 gene in bone marrow hematopoietic cells were detected.
Results: (1) mdr1
gene was successfully transferred into murine marrow hematopoietic
cells(the transfection rate was 35%) and the murine model of scheduled bone
marrow transplantation was successfully established; (2) The proper lethal
dose of 60Co-¦Ã rays irradiation was 1.5Gy; (3) The optimal
infusion time of transferred hematopoietic cells was 24h and 48h
twice point after irradiation.
Conclusion: A stable and efficient method for establishing
the scheduled bone marrow transplantation murine model is set up.
