Objective: Observing the effects of shuang-huang-lian (双黄连) on the murine brain tissue infected with CVB3.
Methods: 1.The primary nerve cells taken from the brain of new born mice were made to culture, and then, various concentrations of shuang-huang-lian (双黄连) added for measuring the LD50 of cells (MTT method). 2. 70 BALB/C mice divided into 3 groups: In groups A and B, the mice were initially infected intracranially with CVB3 (106,105,and 104 PFU CV3 were respectively injected), then groups A and B were injected peritoneally with Hanks or shuang-huang-lian (双黄连) per day respectively. The group C is as control. 3. Statistical analysis: Analysis of chi-square (X2 test) and variance( F test )were used.
Results: 1. The LD50 of shuang huang lian (双黄连)on nerve cells is 0.065mg/ml. 2. The number of dead mice in group B (9/30) were decreased significally less than that in group A (16/30) (P<0.05). But there was no statistical significance among the small groups while comparing in the same titer of CVB3 infected, may be because the number of mice were small.
Conclusion: The preliminary experiments demonstrated that shuang huang lian (双黄连)may have no the effects on the mice brain tissue infected with CVB3.
