Text Box: CHANGES OF GENE EXPRESSION PROFILE IN EONATAL RATS WITH HYPOXIC-ISCHEMIC BRAIN DAMAGE

Objective: To study the changes of gene expression profile in hypoxic-ischemic brain damage (HIBD).
Methods: HIBD was induced by a combination of unilateral carotic artery ligation and transient hypoxia with inhalation of 8% oxygen.Twenty neonatal rats were randomly divided into HIBD group (n=10) and normal control group (n=10).Brain tissue was obtained from HIBD rats at 6 hour after HI and normal rats in same time. Extraction of mRNA and probe preparation was completed and hybridized to gene chip (2048 points). Hybridization signals were detected using fluorescent image.
Results: Gene expression profile at 6 hour after HI was different between HIBD and normal rats.47 genes had overexpressions and 56 genes showed decreasing expression in HIBD group comparing with normal group. NF-kB activator, neural cell adhesion protein mRNA and apoptosis susceptibility protein mRNA had overexpression in HIBD group, ratios were 10~350.However,apoptosis protein inhibitor showed decreasing expression in HIBD group, ratio was 0.179.
Conclusions: These results suggested gene expression profile showed changes in HIBD. Apoptosis genes had overexpression and anti-apoptosis genes had decreasing expression.
