GENE EXPRESSION OF MMP-3 IN PULMONARY VASCULARREMODELING OF RATS
WITH PULMONARY HYPERTENSION INDUCED BY MONOCROTALINE Wang DW, Qin YM Nanjing Children's Hospital,
Nanjing, China Objective: To explore
the etiology of pulmonary hypertension with relation to the pulmonary
expression of metalloprotease3 (MMP-3). Methods: The model of
rat with pulmonary hypertension was induced by monocrotaline (MCT)
subcutaneous injection and the pulmonary arterial pressure was measured by
cathetrization and the pulmonary MMP-3mRNA expression levels were determined
by RT-PCR The contents of hydroxyproline in main pulmonary artery were
tested by using chromometry. Results: The
pulmonary arterial pressure was increased significantly on day 21 after
monocrotaline injected and come to the peak on day 28 in the study The
expression level of MMP-3 m RNA in pulmonary was increased significantly on
day 7 and then come down gradually on day 14、21、28.The content of hydroxyproline n main
pulmonary artery was decreased on day 14 compared with control then was
increased on day 21、28. Conclusion: MMP-3
over-expression involved in the development of pulmonary hypertension and
might play the role of target during earlier stage of pulmonary arterial
remodeling.
